Wnt糖蛋白是存在于多種生物體內(nèi)的一種細(xì)胞外配體,Wnt作為形態(tài)發(fā)生素通過(guò)激發(fā)細(xì)胞內(nèi)遠(yuǎn)離信號(hào)發(fā)送區(qū)域的濃度依賴(lài)反應(yīng)控制胚胎形態(tài)發(fā)育。Wnt通路調(diào)節(jié)動(dòng)物發(fā)育過(guò)程中的多個(gè)重要環(huán)節(jié),例如細(xì)胞增殖、細(xì)胞遷徙及細(xì)胞分化。Wnt蛋白通過(guò)自分泌或旁分泌作用與位于細(xì)胞膜上的受體相結(jié)合,激活細(xì)胞內(nèi)信號(hào)通路調(diào)節(jié)靶基因的表達(dá),對(duì)細(xì)胞的增殖、分化、遷移、極性化和凋亡起到重要作用。

The predicted size of mouse WNT-5a is a monomeric protein containing 356 amino acid residues. Due to glycosylation, it migrates at an apparent molecular weight of ~45 kDa by SDS-PAGE analysis under non-reducing conditions. StemRD’s product is expressed from a mouse cell line, and purified with a proprietary process that is distinct from the published method. ? Purity: Greater than 85% as determined by SDS-PAGE and HPLC analysis ? Biological Activity: The activity was determined by using a TCF reporter gene assay in 293 cells co-transfected with Frizzled-4 and LRP-5. WNT-5a activates (instead of inhibits) the TCF reporter gene in this assay (Milkels AJ, et al., PLoS Biol, 4:e115, 2006). This activation mode is utilized because activation assays are generally more reliable than inhibition assays, as they are less prone to any non-specific inhibitory contamination in the preparation.
